The Marasco Lab offers exciting research opportunities in human antibody engineering and regenerative medicine. With our numerous tools including state-of-the-art humanized mouse models, we are making discoveries in B cell development, host immunity and vaccine design for both infectious disease and cancer immunotherapy applications. Our research team focuses on identification of clinically relevant human monoclonal antibodies and studies of their mechanism of action. Our humanized mouse models are also being used to investigate the pluripotency of adult stem cells with applications in the field of regenerative medicine.
Working in Dr. Marasco's laboratory provides exposure to the most recent developments in the field of antibody engineering, humanized mouse models and adult stem cell biology.

Lab Manager (ID 2016-4683)

The primary role of the Manager is to ensure a safe and efficient laboratory environment. This candidate must have laboratory management skills and take responsibility for the daily operation of the research lab, including equipment, supplies, software, and documentation. Laboratory management skills include:
- Maintaining and operating the lab’s major equipment. The Laboratory Manager is to act as a technical expert/advisor on all equipment issues. This individual is responsible for training lab members on the correct use of lab equipment, evaluating and recommending new technologies, and assisting the administrative team with equipment budgets and potential equipment purchases. S/he is also responsible for planning, coordinating and directing the operations of all equipment for effective maintenance and repair programs and service contracts.
- Ordering, organizing, and restocking core lab supplies. The job requires communicating with representatives of companies marketing laboratory and scientific products and arranging for demos and/or group discounts when necessary.
- Abilities to maintain a safe, secure yet functional lab environment are a must. The manager will be responsible for organizing and allocating the space in the lab’s refrigerators, freezers (-20˚C, -80˚C and liquid N2), and CO2 incubators as well as organizing periodical lab cleaning up. S/he will also play a central role in distributing other lab maintenance duties to technicians, fellows and/or scientists.
- Responsible for archiving bacteria glycerol stocks and plasmids, and electronically cataloging them in a computer-based laboratory database, and helping to locate archived reagents for all lab members. In addition, s/he will be responsible for creating new database archives for liquid N2 stored cells. The individual will perform reagent preparation and shipping/handling related to incoming and outgoing Material Transfer Agreements (MTAs). This includes preparation of electronic plasmid maps and QC the materials into and out of the lab
Scientifically, the individual will be responsible for creating and maintaining immunoglobulin gene databases for the research team. In this role, s/he will be responsible for analyzing immunoglobulin gene sequences that are generated in the lab and compiling amino acid sequence tables, alignments and gene family assignments for publication, including performing web based searches of gene and protein structures and functions, performing web mining, alignments, etc. As a result, the position requires a detailed understanding and working knowledge of all aspects of bioinformatics as described in the “The Genome Access Course” by Cold-Spring Harbor Laboratory's Genome Research Center, and the ability to apply this knowledge as support to ongoing laboratory projects.
The individual will be an integral member of the laboratory and is expected to make presentations at lab meetings. His/her intellectual input and co-authorship are encouraged.
Skills and Qualifications:
- Advanced degree in biomedical sciences or related field with a MS/PhD degree in molecular biology and immunology is preferred. Bachelor's required.
- The preferred candidate will have a minimum of 3 years of experience. S/he must have previous experience managing a laboratory setting, including equipment, and be able to oversee laboratory operations immediately.
- Programming skills to manage robotics and an array of equipment is also preferred – including but not limited to an automated system that consists of a Tecan liquid handler, colony picker, two power washers, two incubators and a MSD reader; a flow cytometry analyzer and a sorter; a BiaCore T100 and an OctRed 96 instrument, a multi-label plate reader and miscellaneous routine equipment.
- Proficient computer skills are also expected with competency in Word, Excel, etc with use of both PC and Mac.
- The applicant must be a self-starter with the ability to work well within a fast-growing team environment and be able to work on his/her own with minimal daily supervision.
- The individual is expected to be able to multi-task and be well organized, execute good time management skills and pay careful attention to details, as well as have excellent communication skills.
Senior Research Animal Technician (ID 2016-4979)
The Marasco lab is seeking a highly motivated, bright individual to join its research team as a Senior Research Technician who will be primarily involved in small animal experiments that are critical to ongoing research projects in the lab.
Responsibilities include but are not limited to performing breeding colony management, injections (IV, IP, SC, IM, ID), blood sampling techniques (sub-mandibular and cardiac puncture), necropsy, immunization, tumor measurements, and gamma irradiation for research animals.
S/he will be constructing and maintaining specialized "humanized" mice developed in-house. As such, experiences in tissue procurement and processing including stem cell purification, small animal survival surgery, and isolation/characterization of human and mouse lymphocytes are definite plus. A basic knowledge in molecular and cellular biology and immunology is desired as this person will be expected to contribute significantly towards developing research projects and performing immunoassays with minimal supervision. Other duties include:
- Assist, train or supervise other lab members on animal experiments.
- Perform standard molecular biology assays, cell culture and flow cytometry.
- Document, compile, and assist in interpreting experimental data. Report data to supervisor in oral and written reports and give presentation at group meetings. Construct figures for grant proposals, progress reports and manuscripts.
- Independently operates lab equipment, prepares and orders necessary supply stocks. Shares lab maintenance duties with other lab members.
Skills and Qualifications:
- Bachelor’s degree or higher in biological sciences or related field.
- Self starter, highly motivated team player and ability to multi-tasking
- Proficiency in basic lab techniques and principles of molecular biology and immunology required
- 2-3 years of experience in animal handling while working in a biomedical research laboratory is preferred
- Proficiency in the use of various scientific databases and publications is required.
- Excellent written and oral communication skills as well as strong organizational skills required.
Research Fellows (ID 2016-5087)
Two Positions are available for Research Fellows to join the Marasco Lab's exciting therapeutic antibody discovery and chimeric antigen receptor (CAR) T-cell cancer therapy program.
The fellows will work on different cancer targets and isolate antibodies through antibody panning/selection to identify new human antibodies against each specific cancer target including affinity ranking, and in vitro antibody characterization. S/he will then generate cancer target specific CAR constructs, create CAR T-cells, and carry out in vitro assays to test their biological activity. The fellows are expected to set up appropriate tumor model(s) for their perspective project in humanized mice and perform in vivo experiments to evaluate therapeutic potential of specific CAR T-cells for a given cancer target.
The candidate must be a self-starter and highly motivated team player. S/he is expected to work independently, well versed in the related literature, and contribute intellectually to his/her research program. S/he will design and construct new yeast antibody display libraries when necessary.
Skills and Qualifications:
Ph.D. degree with documented research experience in molecular biology and immunology, especially in antibody discovery and/or T cell therapy. Experiences in cancer animal model and relevant molecular, biochemical and immunological techniques are preferred. We are particular interested in the candidates with prior research experience in the area of hematologic malignancies and CNS/brain tumors. The individual is also expected to be able to parallel processing (good time management skills), well organized, and pay careful attention to experimental details. While supervisory experience will be a plus, the candidate should expect to be fully involved in hands-on bench work.
